RNA pathways to cognition
Our Research
Our mission is to understand the molecular basis of psychiatric disorders that increase the risk of suicide.
We are using a reverse translational approach to elucidate the pathophysiology of psychiatric disorders. Initially we investigate gene expression in postmortem brain from cohorts of individuals with schizophrenia, bipolar disorder and major depression, including cases of suicide. Our recent work revealed that an epitranscriptomic process known as RNA editing, is elevated in the dorsolateral prefrontal cortex (DLPFC) of depressed suicides. Following these studies of human brain, we are testing the effects of stress on mouse behavior and neurochemistry. RNA editing regulates brain development and synaptic plasticity, which are impaired in psychiatric disorders, including schizophrenia and depression. Our results suggest that prenatal stress triggers abnormalities in the glutamate system and in RNA editing, which may be precursors for psychiatric illness. We have consistently identified sex differences in these data. The objective of this research is to identify new targets for improved therapeutic interventions and to discover biomarkers with which we can predict suicidal behavior, so that preventative strategies can be developed.
Training Opportunities in the Sodhi Lab:
Students, fellows or residents who join Dr. Sodhi’s lab can expect to receive training in molecular genetics, gene expression studies of rodent or human tissue, behavioral pharmacology, mammalian cell culture, in addition to gaining expertize in the broader area of the neuropharmacology of schizophrenia, mood disorders and suicide.
Descriptions of some of our projects are below.
Epitranscriptomic basis of resilience to psychological stress in mice and humans.
understanding the molecular basis of cognition and emotion
The overall goal of this project is to understand the molecular pathways that increase resilience to stressful life events. It is established that mental illness runs in families (genetics) and that mental illness is triggered or exacerbated by environmental stress. We are testing the effects of psychological stress on behavior, and on the expression of genes in specific brain regions that are important in learning, memory and anxiety behaviors.
Sex differences in major depression and suicide
Studies of glutamatergic pathways
Gene expression analyses reveal that female patients with depression may have elevated expression of glutamate receptor genes in the dorsolateral prefrontal cortex. This region is important for executive function and implicated in response to antidepressant therapies including TMS. Both males and females who died by suicide appeared to have increased expression of specific glutamatergic genes: the genes encoding GluN2B (GRIN2B), GluK3 (GRIK3) and mGluR2 (GRM2) and specific glutamate transporters. These data indicate that the glutamatergic system may contain many targets for antidepressant drug development. The glutamate synapse is illustrated on the left.
Glutamatergic genes in schizophrenia
Studies of the glutamate system in the thalamocortical circuitry
Accumulating data support the glutamate hypothesis of schizophrenia. Our investigations have led to the discovery of reduced expression of glutamatergic genes in laser-captured relay neurons of the medial dorsal thalamus (pictured on the right). These results indicate that the thalamocortical circuitry is disrupted in schizophrenia, perhaps underpinning the disorganized thought exhibited by schizophrenia patients.
Neuronal morphology predicts mental health
Dendritic branching, spine density and morphology are measures of the number, strength, and stability of synaptic contacts. These morphological features are predictors of cognitive function and mental health. We are measuring variations in neuronal morphology in mice exposed to psychological stress. The neuron pictured is from the CA1 region of the mouse hippocampus.
Identification of predictors of suicide risk.
Analyses of psychiatric patient populations
The overall goal of this project is to develop an objective method to assess suicide risk. The rate of suicide has steadily increased over the past decade, with over 44,000 deaths per year. Large epidemiological studies have identified a number of risk factors for suicidal behavior in adults and key amongst them are preexisting mental illnesses, particularly mood disorders and depression, impulsivity, hopelessness, anhedonia and insufficient coping strategies during stressful events. Tragically, many patients do not report that they are suicidal and therefore fail to receive appropriate treatment. We could reduce the frequency of suicide if we had an objective means to identify patients who are at high risk, so that these patients are targeted for immediate and intensive psychiatric treatment. In this project, we plan to develop a blood test for suicide risk, including genetic and inflammatory biomarkers.

Call to Action: Neuroscience Research
The brain is the final frontier of the life sciences. Investigating the brain in health and disease is where you can have the most impact. The questions are profound, the illnesses are fascinating, and we finally have tools with which we can make unprecedented progress. Although mental illness is the leading cause of disability and death globally, we still lack effective treatments for psychiatric and neurological disorders. This is an invitation to join us in our work to improve our understanding of the molecular basis of mental illness!
